MPTP所致帕金森病模型黑质区TNF-α、IL-1β的表达及灵芝孢子油的调控作用

【摘要】 目的研究1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)所致帕金森病模型中脑黑质区的免疫应答情况,探讨灵芝孢子油对MPTP引起的神经炎症反应的调节作用。方法C57BL小鼠分为MPTP组(组1)、灵芝孢子油+MPTP组(组2)、正常对照组(组3)。组1皮下注射MPTP30mg/kg,连续6d。组2在注射MPTP前两天开始鼻饲灵芝孢子油每日1.5g/kg,连续8d。于末次注射MPTP后第8天用Western免疫印迹法检测中脑腹侧部肿瘤坏死因子-α(TNF-α)蛋白含量;用荧光半定量RT-PCR法检测中脑腹侧部TNF-α、白细胞介素-1β(IL-1β)mRNA表达量。结果①TNF-α蛋白的相对含量:组1比组3明显增多,组2比组3略高,三组组间比较有明显差异(P<0.01)。②TNF-αmRNA、IL-1βmRNA的表达量:组1比组3明显增高,组2比组3减少,组1与组2比较有显著性差异。结论MPTP所致帕金森病模型黑质区炎症因子表达明显增高,灵芝孢子油能有效下调MPTP引起的神经炎症反应。

【Abstract】 Objective To investigate the immune response of the substantia nigra in the MPTP-induced model of Parkinson’s disease(PD), and explore the regulation of ganoderma spore oil on the inflammatory response induced by MPTP. Methods C57BL mice were randomly assigned to three groups. Group 1 mice were hypodermically injected with MPTP (30 mg/kg·d) for 6 days. Group 2 mice were administered with ganoderma spores oil by nasogastric feeding (1.5 g/kg·d) for 2 days. Two days later, MPTP (30 mg/kg·d) together with ganoderma spores oil (1.5 g/kg·d) were given to the group 2 mice for 6 days. Group 3 (control group) was treated with normal saline. At the 8th day of MPTP injection, real-time PCR and Western immunoblot were used to detect the mRNA expressions of TNF-α and IL-1β and the protein content of TNF-α in the mesencephalon ventral part. Results The protein content of TNF-α in group 1 was significantly higher than that in group 2 and the control group. The mRNA levels of TNF-α and IL-1β in group 1 increased while the levels in group 2 decreased compared with the control group. There were significant differences in the mRNA levels of TNF-α and IL-1β between group 1 and group 2. Conclusions The expression of pre-inflammatory cytokines in the substantia nigra increased significantly in mice with MPTP-induced PD. Ganoderma spore oil may attenuate the neuroinflammatory response by reducing the generation of pro-inflammatory cytokines.
