不同方法对枣叶片总DNA提取效果的影响
Effect of different methods on Chinese jujube DNA extraction from leaves
【摘要】 以串杆枣的成熟鲜叶为试材,分别采用-20℃、-70℃和硅胶脱水干燥3种方法保存样品,采用简易CTAB法、高盐沉淀法与高盐低pH值法分别对3种方法保存的样品及对照样(鲜叶)进行了基因组DNA提取效果的比较分析。结果表明,3种方法均能有效地从枣叶片中获得质量较高的DNA,以高盐沉淀法所得的DNA纯度最高,高盐低pH值法次之。就DNA的产量而言,高盐沉淀法的DNA产量相对较低,低于其他的2种方法,但未达到显著水平。硅胶干燥法和-70℃超低温保存的样品与鲜叶相比,所提DNA的质量和产量均无明显差异,均能满足PCR扩增要求。硅胶保存法适于野外远距离采样。
【Abstract】 Experiment was conducted with mature leaves of Zizyphus jujuba Mill. cv. Chuangan (stored at -20 ℃,-70 ℃,dried with silica gel) and 3 methods,including CTAB,high salt precipitation and high salt and low pH were used for total DNA extraction for studying their effects on DNA extraction. The results showed that high-quality DNA can be obtained with all the methods. The purity of DNA extracted with high salt precipitation method was the best,and then the low pH method. But the yield of DNA with high salt precipitation method was the lowest. The purity and yields of DNA extracted from the leaves preserved in silica or -70 ℃ were not significantly different from fresh leaves. Storing in silica gel was suitable for sampling from open country or far away place.
