复方丹参对大鼠急性脊髓损伤后核因子-κB活性的影响

【摘要】 目的探讨复方丹参对于大鼠急性脊髓损伤后核因子-κB(NF-κB)活性的影响。方法成年Wistar大鼠36只,体重200~250g,雌雄不限,随机分为3组,正常组4只,脊髓损伤后复方丹参处理组(A)和生理盐水对照组(B)各16只,损伤后不同时间点(12h、1d、3d、5d)处死大鼠,用苏木精-伊红(HE)染色观察脊髓组织病理变化,用免疫组化染色检测NF-κB阳性细胞。免疫印记分析法,检测细胞内NF-κB的活性变化。结果HE染色发现脊髓组织病理学改变A组轻于B组。免疫组化免疫印记分析A、B2组均发现NF-κB表达,且NF-κB表达为B组大于A组。结论急性创伤性脊髓损伤后,复方丹参可以抑制NF-κB的活性表达,对减轻NF-κB介导的炎性反应起到有益作用。

【Abstract】 Objective To investigate the effect of salvia miltiorrhiza on nuclear factor-kappaB (NF-κB) activity after spinal cord injury in rats.Methods Adult Wistar rats were randomly divided into three groups: group A with spinal cord injury caused by Allen’s weight drop and treated with salvia miltiorrhiza,group B treated with normal saline group and the normal group.The rats were sacrificed at 12 hours and 1,3,5 days after surgery,and then the specimen sections were stained with hematoxylin and eosin,and immunohistochemistry.The injured spinal cord tissue was observed.At the same time the activity of NF-κB was detected using western blot analysis in the injured spinal cord.Results The histopathology changes in spinal cord tissue observed with hematoxylin-eosin staining microscopic examination in group B were worse than those in group A.Expression of NF-κB were observed in the two groups,and the expression of NF-κB on group B was higher than that in group A (P<0.01). Conclusion Salvia miltiorrhiza could inhibit expression of NF-κB after spinal cord injury in rats.
