钌(Ⅱ)配合物与DNA相互作用的光谱研究
Spectra studies of ruthenium (Ⅱ) polypyridyl complex with DNA
【摘要】 目的设计合成一个钌(Ⅱ)多吡啶配合物[Ru(dmp)2(dpip)]2+(dmp=1,10-邻菲咯啉,dpip=2-(6,9-二氧环己环苯基)咪唑并[4,5-f]邻菲咯啉),研究该配合物与DNA的相互作用。方法采用元素分析、质谱和1H NMR对其进行表征,用电子吸收光谱、黏度测试、DNA熔点变化和圆二色谱研究配合物与DNA的作用。结果与结论所合成的配合物结构得到确证,该配合物与DNA之间通过插入作用结合。
【Abstract】 Ruthenium(Ⅱ) polypyridyl complex [Ru(dmp)2(dpip)]2+(dpip=2-(6,9-dioxanophenyl)imidazophenanthroline,dmp=2,9-dimethyl-1,10phenanthroline) has been synthesized and characterized by element analysis,ESMS and 1H NMR.The interactions of the complex with DNA were investigated by electronic absorption spectra,thermal denaturation,viscosity measurement and circular dichroism.The results show that the complex [Ru(dmp)2(dpip)]2+ intercalates into the DNA base pair via the intercalative ligand.
