神经再生相关营养因子的提取及生物学特性检测

【摘要】 目的提取神经再生微环境中的神经再生营养活性物质。方法取York猪暴露坐骨神经,截断后缝接入硅胶管中,形成神经再生室,吸取硅胶管内液体,凝胶柱层析分离纯化神经再生液,雪旺细胞(SC)增殖实验检测所提取的神经再生营养因子的活性。结果凝胶层析分离神经再生室内液体可得到2个峰,经检测2号蛋白峰可以促进SC增殖。结论神经再生液中的提取物具有神经营养功能,可用于周围神经修复。

【Abstract】 Objective To isolate nerve regeneration-related neurotrophic factor (NRNF) from nerve regenerative microenviroment. Methods The sciatic nerve of York swine was desected and bridged by silicon tube to form nerve regeneration chamber. We take solution from silicon tube, and then isolate and purify the nerve regeneration solution by gel chromatography, and detect NRNF physiological activity by schwann cell (SC) proliferation test. Results Two protein peaks (No.1, No.2) were harvested from gel chromatography and No.2 protein peak can promote SC proliferation. Conclusion The NRNF from nerve regenerative microenviroment have the nerve neurotrophic function, can promote the nerve regeneration.
