口腔鳞癌细胞FasL表达对肿瘤浸润淋巴细胞及癌细胞凋亡的影响
Effect of Expression of FasL in Oral Squamous Cell Carcinoma on the Apoptosis of Carcinoma Cells and Tumor Infiltrating Lymphocytes
【摘要】 目的研究口腔鳞癌细胞FasL表达及其对癌细胞和肿瘤浸润淋巴细胞(TIL)凋亡的影响。方法38例口腔鳞癌为实验组,10例正常口腔黏膜为对照组。采用免疫组织化学方法和末端脱氧核糖核酸转移酶介导的原位末端标记技术,观察癌组织FasL表达,计算TIL和癌细胞的凋亡指数,观察癌细胞FasL表达与TIL及癌细胞凋亡的关系。结果FasL在正常口腔黏膜不表达,在鳞癌组织中表达明显上调(P<0.05),FasL表达与口腔鳞癌分化程度无关(P>0.05)。口腔鳞癌细胞FasL表达阳性组癌细胞凋亡指数明显低于FasL表达阴性组,同时TIL的凋亡指数高于FasL表达阴性组(P<0.05),口腔鳞癌细胞凋亡指数与TIL凋亡指数呈负相关(r=-0.72,P<0.05)。结论口腔鳞癌细胞可能通过上调表达FasL诱导TIL的凋亡,间接地使癌细胞凋亡下调而保护了自己,是口腔鳞癌组织免疫反攻击的体现。
【Abstract】 Objective To study the effect of expression of FasL in oral squamous cell carcinoma(OSCC) on the apoptosis of carcinoma cells and tumor infiltrating lymphocytes(TIL).Methods The expression of FasL protein in OSCC cells and apoptosis of tumor cells and TIL were detected by immunohistochemical method and in situ terminal deoxynucleotide transferase labeling technique(TUNEL) respectively in 38 cases of OSCC.The relationship between the expression of FasL and apoptosis in tumor cells and TIL was obiserved.Results There was no expression of FasL in oral normal mucosa and an apparent up-regulated expression in OSCC(P<0.05).The FasL expression was not related to the differentiation degree of OSCC(P>0.05).The apoptotic index(AI) of tumor cells in FasL positive OSCC was remarkably lower than that in countpart.The AI of TIL in FasL positive OSCC was higher than that in FasL negative OSCC(P<0.05).The AI of tumor cells was reversely correlated with that of TIL((r=-0.72),P<0.05).Conclusion OSCC cells may induce the apoptosis of TIL and down-regulate their own apoptosis indirectly through up-regulating their expression of FasL.Therefor,the up-legulated expression of FasL in OSCC may protect itself and is an implication of immune counterattack.
