【摘要】 目的观察槐耳清膏在体外对人肝癌细胞株HepG-2凋亡的影响,为槐耳清膏在临床上治疗肝肿瘤提供理论依据。方法采用MTT法、TUNEL法及流式细胞术,观察不同浓度的槐耳清膏对体外培养的人肝癌细胞株HepG-2的生长活力、细胞凋亡、DNA合成能力的影响;琼脂糖凝胶电泳检测DNA-LADDER的形成。结果:槐耳清膏在体外能抑制人肝癌细胞HepG2的生长,并具有时间及剂量依赖性;发生作用后,细胞生长有明显的凋亡特征性改变;细胞DNA的合成能力下降。结论:槐耳清膏能有效地抑制体外肝癌细胞生长,其机制可能与抑制肝癌细胞DNA合成及诱导肝癌细胞凋亡有关。更多还原

【Abstract】 Objective To study the effect of Tramets robinophila on the apoptosis of the human hepatocellular carcinoma cells line HepG-2 and provide the experimental basis for Tramets robinophila treating liver cancer in clinic.Methods The effects of Tramets robinophila on proliferation,apoptosis and DNA synthesis of HepG-2 cells were determined by MTT assay,TUNEL,and flow cytometry.Agrose electrophoresis was applied to investigate DNA-LADDER.Results The growth of HepG-2 cells was inhibited by Tramets robinophila in vitro in a time/dose-dependent manner.Tramets robinophila induced apoptosis of HepG-2 cells and reduced their DNA synthesis.Conclusion Extractum trametes robiniophila murr is effective in inhibiting growth of HCC in vitro.The mechanism of antineoplastic effect of trametes robiniophila might be inhibition of DNA synthesis and induction of apoptosis of tumor cells.更多还原
