过量维甲酸对金黄地鼠胚胎神经管维甲酸受体β表达的影响

【摘要】 目的探讨过量维甲酸(RA)对金黄地鼠胚胎神经管维甲酸受体(RARβ)基因和蛋白表达的影响。方法采用原位杂交和免疫组织化学方法及图像分析,半定量分析金黄地鼠胚胎神经管正常发生及RA致神经管畸形过程中RARβ转录和表达水平的变化。结果正常对照组RARβ表达水平随胚龄增加呈先降低后升高并再次减弱的趋势。实验组在灌服过量RA后3、6h,RARβ表达水平明显低于正常对照组(P<0.01);14h时表达升高显著高于正常对照组(P<0.05);22h时其表达再次降低明显低于对照组(P<0.01);46、96h表达逐渐增强,96h时明显高于对照组(P<0.01)。结论过量RA引起的RARβ转录和表达水平的变化与RA致金黄地鼠神经管畸形相关。

【Abstract】 Objective To investigate the effect of excess retinoic acid(RA)on the expression of Retinoic acid receptor-beta(RARβ)in neural tube of golden hamsters.Methods Through in situ hybridization,immunohistochemistry and image analysis methods,the difference of RARβ expression between normal and defected neural tube induced by excess RA was analyzed in golden hamsters.Results Expression of RARβ in the normal embryonic neural tube tissues decreased at first,then increased,but at last decreased with the age of the embryo.3 to 6 hours after feeding with retinoic acid,the expression level of RARβ were significantly lower than that in the corresponding control groups(P<0.01).Then 14 hours after feeding,expression level was significantly higher than that of the corresponding control group(P<0.05).22 hours after feeding,expression level significantly lower than that of the corresponding control group again(P<0.01);46 to 96 hours after feeding,their expressions increased gradually,and 96 hours after feeding,the level was higher than that of the corresponding control group significantly(P<0.01).Conclusion The changes of RARβ expression induced by excess RA are related to the neural tube defects in golden hamster.
