甘露糖受体在小鼠精子中的表达及其在受精中的作用

【摘要】 为研究小鼠精子甘露糖受体的表达及其对受精的影响,采用FITC-DMA作为探针,对精子甘露糖受体(MR)进行定位,获能精子分别用DMA、D-甘露糖处理后和卵子共培养;或卵子分别用甘露糖苷酶、ConA处理后与获能精子共培养,在相差显微镜下计数各组卵子的精子结合数。结果显示小鼠精子头部呈现2种荧光标记:①整个精子头部有荧光标记;②赤道后区有荧光标记。DMA、D-甘露糖、甘露糖苷酶、ConA对精子-透明带结合有显著抑制作用,而对精子-卵膜结合没有影响。说明甘露糖受体在精子-透明带相互作用中具有调节作用。

【Abstract】 To investgate the expression and localization of mannose receptor in mouse sperm plasmalemma and its role in the mouse fertilization.D-mannose binding sites or mannose receptors were identified using α-D-Mannosylated-FITC-albumin.Capacitated spermatozoon treated with DMA and D-mannose respectively was incubated together with ovum,or ovum treated with mannosidase and ConA respectively was incubated together with capacitated spermatozoon,then the number of spermatozoa bound per zona pellucida was calculated.The results showed that two main fluorescent patterns were seen at the head level,one covering the entire head and the other with the label concentrated at the equatorial segment,and their expression did not chang with capacition and acrosome reaction.Incubation of capacitated spermatozoon in the presence of DMA and D-mannose resulted in decrease in the number of spermatozoon bound per zona pellucida,and this also occurred when zona pellucida-onclosed egg treated with mannosidase and ConA.DMA,D-mannose and mannosidase had no effects on spermatozoon bound per ovum plasmalemma,but ConA increased the bound significantly.Conclusion:Mannose receptor may have a role in the interaction between sperm-ZP.
