氡染毒大鼠支气管肺泡灌洗液蛋白质的双向电泳分析

【摘要】 观察氡染毒后大鼠支气管肺泡灌洗液(Bronchoalveolar lavage fluid,BALF)蛋白质表达谱的变化。雄性Wistar大鼠吸入氡的累积剂量达200工作水平月(Working level month,WLM)后,收集大鼠BALF,提取BALF细胞总蛋白,采用双向电泳技术(Two-dimensionalel ectrophoresis,2-DE)分离总蛋白质,银染显色,进行差异蛋白质的分析。ImageMaster 2D Platinum软件分析结果显示正常组和氡染毒组大鼠BALF细胞总蛋白双向电泳图谱上分别可见到645和554个蛋白点,共获得48个显著的蛋白质差异点,其中23个点在正常大鼠BALF中表达或表达明显增强,25个点在染毒大鼠BALF中表达或表达明显增强。结果提示,正常大鼠与氡染毒大鼠BALF细胞的蛋白质表达谱存在差异。

【Abstract】 In this paper the different protein expression of BALF in rats inhaled radon was analyzed by use of two-dimensional electrophoresis (2-DE). Male Wistar rats were exposed to radon with the cumulative dose up to 200 working level month (WLM), then the bronchoalveolar lavage fluid (BALF) was collected and the total proteins were extracted. 2-DE was employed to isolate the total proteins of BALF. The gels were stained with silver and differential protein expressions were analyzed with Image Master 2D Platinum software. The total protein spots in the gels of the control and radon-inhaled rats were 654 and 554 respectively, containing 48 differential protein spots in which 23 ex-pressed and obviously increased in the control group and 25 expressed and obviously increased in rats radon-inhaled group. The results indicate that the protein expression of BALF between rats radon-inhaled and the control are dif-ferent to some extent.
