影响大肠埃希菌感受态细胞因素的探讨
Study on Impacting Factors of Escherichia coli Competent Cell Preparation
【摘要】 研究不同型号的菌株、不同转化溶液、感受态细胞的存放时间、转化时间及转化后的复苏时间等对质粒转化效率的影响。结果表明,转化溶液浓度和感受态细胞的存放时间对转化效率有明显的影响,而转化时间、冰浴时间和转化后的振荡培养复苏时间对转化效率的影响不明显。
【Abstract】 A number of factors including bacterial strains,transformation solutions,freezing duration and storage duration were evaluated for their effects on transformation ability of competent Escherichia coli cells.The different effects of the transformative condition which included bacterial strains,CaCl2 concentration,of the competent cell status,of the various transformative time and of the incubative time on the transformative plasmid were investigated.The host cell was Escherichia coli DH5α and JM109.The results showed that effects of different transformation solutions and the competent cell status on the transformative frequency were obvious,but the effects of the transformative time,freezing duration and incubative time after transformation were not significant.
