大肠埃希菌质粒DNA提取方法的优选

【摘要】 采用酚/氯仿少量提取法、异丙醇沉淀法、煮沸法以及微波炉法提取细菌质粒,然后分别对各种方法所提取的质粒进行琼脂糖凝胶电泳分析,并用紫外分光光度计测定其OD值,对其优劣性进行全面的分析。结果表明,异丙醇提取法所提取的质粒DNA纯度和产量高(其DNA片段荧光较强,具有清晰的亮带,OD260/OD280约为1.77,DNA样品浓度约为195μg/mL),且重复性好,具有结果稳定的特点,达到了分子生物学试验要求,适合大多数实验室使用。

【Abstract】 In this experimental study,phenol-chloroform extraction method,isopropyl alcohol extraction method,simple and quick boil method,and microwave heating method were used to extract plasmid.Then agarose gel electrophoresis was used to analyze the respective plasmid DNA obtained by different methods.Finally,OD values were measured by the method of ultraviolet spectrophotometer determination and each method’s advantages and disadvantages were analyzed.Good yields with high purity and good repetition were found to have been obtained in the second method(the fragment of DNA has strong fluorescence and clear bright band,OD260/OD280 is 1.77,the density of DNA sample is 195 μg/mL approximately),the results also showed that this method displayed good reproducibility and stronger stability,which may be used for molecular biology experimentation.Therefore it is concluded that the isopropyl alcohol extraction method is very suitable to be used by most laboratories.
