日本血吸虫腺嘌呤磷酸核糖转移酶的生物信息学鉴定与分析

【摘要】 目的鉴定日本血吸虫中存在的腺嘌呤磷酸核糖转移酶,分析其蛋白结构特征。方法采用双向同源比对、结构域搜索和系统发育分析等生物信息学手段,在已有转录组和蛋白质组数据基础上鉴定日本血吸虫的腺嘌呤磷酸核糖转移酶,并采用同源建模方法获得日本血吸虫腺嘌呤磷酸核糖转移酶的结构特征。结果获得了2条腺嘌呤磷酸核糖转移酶的同源蛋白序列;分析了这2条序列的EST丰度、理化性质及蛋白三维结构等信息。结论日本血吸虫至少存在2个腺嘌呤磷酸核糖转移酶的异构体。

【Abstract】 Objective To identify adenine phosphoribosyltransferases in Schistosoma japonicum and analyze their structural features.Methods Based on the accessible transcriptome and proteomic data,the S.japonicum adenine phosphoribosyl transferases were identified using bioinformatics approaches including bi-directional homology comparison,domain search and phylogenetic analysis.Homology modeling was also performed to describe the structural features of the proteins.Results and Conclusion Two homologue sequences of adenine phosphoribosyltransferase were obtained from S.japonicum,and the EST abundance,physico-chemical properties and three-dimensional structures of them were also acquired.
