EGCG抑制肺癌细胞增殖及其机制的研究
Effect of epigallocatechin-3-gallate (EGCG) on proliferation of human lung cancer cell and its mechanism
【摘要】 目的:研究表没食子儿茶素-3-没食子酸酯(EGCG)对人肺癌细胞增殖的影响及机制.方法:培养肺癌细胞A549,加入EGCG制作细胞抑制曲线;EGCG作用于人肺癌A549细胞株前后通过Hoechst染色检测细胞凋亡.RT-PCR及Western Blot印迹法研究survivin的mRNA及蛋白表达的变化.结果:EGCG可明显抑制A549细胞的增殖,60mg/LEGCG作用后,细胞出现明显的凋亡.同时肺癌细胞中sur-vivin mRNA和蛋白表达减少.结论:EGCG可抑制肺癌细胞的增殖,这可能与survivin表达降低有关.
【Abstract】 AIM: To explore the effect of epigallocatechin-3-gallate ( EGCG) on the proliferation of lung cancer cells and its mechanism. METHODS: Human lung cancer cell line A549 was cultured with and without EGCG and its suppressed curve was drawn. Apoptosis of the cells was analyzed by Hoechst staining. The expressions of survivin mRNA and protein were investigated by RT-PCR and Western Blot. RESULTS: EGCG inhibited the proliferation of the lung cancer cell A549 and cell apoptotic rate was increased to 18% when A549 was cultured with 60 mg/L EGCG (P<0.01). The expression of survivin was inhibited significantly in A549 cells treated with 60 mg/L EGCG. CONCLUSION: EGCG can inhibit the growth of human lung cancer cells and induce cell apoptosis. Its mechanism may be associated with blocking survivin expression.
