榄香烯复合瘤苗HSP70-肿瘤肽活性成分的纯化及鉴定

【摘要】 [目的]探讨榄香烯复合瘤苗HSP70-肿瘤肽活性成分的纯化方法。[方法]以小鼠HCa-F肝癌和L615白血病细胞为肿瘤模型,分别制备两种榄香烯复合瘤苗,通过亲和层析法制备瘤苗的HSP70-肿瘤肽活性成分,免疫印迹法鉴定两种HSP70-肿瘤肽及其活性。[结果]两种HSP70-肿瘤肽样品均在70 kD附近呈现清晰的一条HSP70-肿瘤肽蛋白带;从HCa-F瘤苗中获得的HSP70-肿瘤肽产量为(2.34±0.28)mg/109个细胞,该产量约是等细胞数L615瘤苗产量的2倍。[结论]该方法适于HSP70-肿瘤肽活性成分的提取并能保持HSP70-肿瘤肽分子的免疫活性。

【Abstract】 [Objective] To investigate a purified method of the active component on HSP70-tumor peptide extracted from Elemene Combo-Tumor cell vaccine(EC-TCV).[Methods] The two of EC-TCV were prepared with cells of HCa-F hepatoma and L615 leukemia in mice respectively,then the active components on HSP70-tumor peptide were extracted from the EC-TCV with affinity chromato-graphy,and HSP70-tumor peptide and their activities were identified by Western blotting.[Results] There was a clear bend near 70 kD in each sample of HSP70-tumor peptide;the output of HSP70-tumor peptide extracted from 1×109 HCa-F cells was 2.34±0.28 mg,the output is higher than the yield from the equal L615 by twice.[Conclusion] The method is a suitable for the extraction of HSP70-tumor peptide and preservation of the activity of HSP70-tumor peptide.
