常用苦寒药过量使用对胃黏膜屏障功能影响的实验研究

【摘要】 目的:研究过量服用传统认为有伤胃作用的九味苦寒药对大鼠胃黏膜屏障功能的影响,探讨“苦寒药伤胃”之说的实质。方法:分别给正常大鼠与脾胃虚寒模型大鼠大量服用9味苦寒药(黄连、山豆根、黄芩、栀子、苦参、黄柏、大黄、防己龙胆草)连续十天。取血、胃黏膜组织分别检测前列腺素E2(PGE2)、超氧化物歧化酶(SOD)及丙二醛(MDA)的含量。结果:动物血中和胃黏膜中PGE2含量不同程度下降;SOD均有不同程度的提高,同时脂质过氧化产物丙二醛(MDA)含量也明显增加,尤其是脾胃虚寒者胃中MDA含量增加较为突出。结论:传统认为有伤胃作用的几味苦寒药在一定程度上可造成胃黏膜屏障功能的损害,表现为胃黏膜PGE2水平下降、氧自由基体系代谢活跃,胃黏膜脂质过氧化产物堆积。

【Abstract】 Objective:To observe the effection of the barrier function of gastric mucosa after intake excessive amount of the nine bitter and cold medicine that can cause damage to stomach and which were definitely recorded in the indications of Traditional Chinese Medicine Textbook.And discuss essential of the theory—“bitter and cold medicine damaong stock”. Methods:Rats of control and Spleen-Stomach deficient cold pattern group were dividedly fed with great amount of the nine bitter and cold medicine(Huanglian、Shandougeng、Huangqin、Zhizi、Kushen、Huangbo、Dahuang、Fangji、Longdacao)for 10 days.By the end of experiment,the content of PGE2、SOD and MDA was separately measured in blood and gatric tissue. Results:In blood and gastric tissue of animals ,the content of PGE2 decreased in some degree,and the content of SOD and MDA increased also,the latter obviously,especially,the content of MDA in the Spleen-Stomach deficient cold pattern group enhanced magnificently.Conclusion:The nine bitter and cold medicine that can cause damage to stomach and which were definitely recorded in the indications of Traditional Chinese Medicine Textbook can make the barrier function of gastric mucosa to damage,and the results of the PGE2 level in the gastric mucosa declining,the oxygen-derived free radicals system metabolizing lively and the products of lipid peroxidation of gastric mucosa stacking were observed.
