利用简易生物反应器大量生产玉花兰根状茎及绿芽

【摘要】 以玉花兰的根状茎为外植体比较了在固态培养基培养、液态培养基振荡培养和生物反应器培养条件下根状茎增殖和绿芽分化情况。结果表明:在生物反应器中根状茎和绿芽分化效果最佳,固态培养次之,液态震荡培养不利于根状茎的增殖和绿芽的分化。通过调查生物反应器中接种密度、空气注入量的影响,发现3L柱型气升式生物反应器内根状茎增殖和绿芽分化的最适接种量为200个外植体,培养8周后每个反应器可获得1120.0个和1779.8个健壮的根状茎和绿芽;反应器中通气量为0.1L/(L.min)时促进根状茎和绿芽分化。

【Abstract】 Rhizome multiplication and shoot differentiation were studied under different culture conditions using the rhizomes of Cymbidum niveo-maginatum as explants.Result shows that the effect of rhizome multiplication and shoot differentiation for bioreactor culture are better than that on solid-agar medium,and liquid shake culture is unsuitable for rhizome multiplication and shoot differentiation.The optimum inoculation density in a 3-L column-type airlift bioreactor is 200 explants in terms of the effects of inoculation density and air injection quantity,and 1 120.0 rhizomes and 1 779.8 shoots in a bioreactor were obtained after 8 weeks of culture.In addition,0.1 L/(L·min)air volume in a bioreactor is optimal for mass propagation of C.niveo-maginatum.
