基于有机-无机杂化材料和铂电极的葡萄糖氧化酶电极的研究
Amperometric Glucose Biosensors Based on Platinum Electrode and Organic-Inorganic Hybrid Material
【摘要】 在制备由聚乙烯醇和二氧化硅溶胶凝胶掺杂而成的有机-无机杂化材料的基础上,将葡萄糖氧化酶用此杂化材料包埋固定在铂盘电极上,利用修饰电极对过氧化氢的电催化还原特性,制得了高性能的安培型葡萄糖氧化酶电极.此酶电极可以在0.05V(vsSCE)下对葡萄糖进行测定,有效地消除了抗坏血酸和尿素的干扰.最适pH值为7.0,载酶量为6%.在选定的工作条件下,此酶电极的灵敏度和线性范围分别为211nA/mmol.L-1和0.1~4.0mmol.L-1,平均响应时间为2.8s左右.
【Abstract】 An organic-inorganic hybrid material based on sol-g el derived SiO2 and PVA has been obtained.Glucose oxidase is immobilized by thi s hybrid material on platinum electrode surface which can catalyze the reduction of hydrogen peroxide and an amperometric glucose biosensor of high performance has been fabricated.The developed sensor is applied to detect glucose with oper ating potential of 0.05 V(vs SCE),which eliminates the interferences of ascorbi c acid and uric acid effectively.Optimum pH is 7.0 and enzymes loading is 6%.U nder selective conditions the biosensor shows excellent sensitivity of 211 nA/mmol/L and a linear range of 0.1~4.0 mmol.L-1.The average response ti me is 2.8 s.
