川芎嗪对缺氧缺血性脑损伤炎症细胞因子作用的实验研究
Research of ligustrazine on brain tissue proinflammatory cytokine in rats after hypoxic-ischemic brain damage
【摘要】 目的观察川芎嗪对新生鼠脑组织缺氧缺血性脑损伤(HIBD)后炎症细胞因子IL-8、TNF-α、IL-10动态变化的影响。方法选用7日龄Wistar大鼠90只,随机分为假手术对照组、HIBD组、川芎嗪治疗组,检测0~48 h 5个不同时间点的效应指标的变化趋势及组间差异。结果HIBD组各时间点IL-8、TNF-α、IL-10浓度均比假手术对照组明显升高(P<0.01),川芎嗪治疗组IL-8、TNF-α、IL-10浓度均比HIBD组降低。结论缺氧缺血早期促炎症细胞因子IL-8、TNF-α和抑炎症细胞因子IL-10表达上升,川芎嗪可减轻缺氧缺血脑损伤后新生鼠脑内炎症程度。
【Abstract】 Objective To study the effects of ligustrazine on the expression of IL-8,TNF-α and IL-10 after hypoxic-ischemic brain damage in rats’ brain tissue.Methods Ninety Wistar rats of 7-day-old were randomly divided into experiment group and control group.The enzyme-linked immunosorbent assay was used to measure mean concentrations of IL-8,TNF-α,IL10 in rats’ brain tissue after the model of HIBD was established.Results The mean concentrations of IL-8,TNF-α,IL-10 in HIBD group were significant higher than those in sham operated group(P<0.01).In ligustrazine intervention group,compared with HIBD group,the concentrations were decreased.Conclusion The concentrations of IL-8,TNF-α and IL-10 were increased after HIBD,ligustrazine could regulate down the inflammation degree,which might be one of mechanisms of ligustrazine treating HIBD.
