激活素受体相互作用蛋白4的基因克隆

【摘要】 目的:克隆新的与激活素受体ⅡA(ActRⅡA)相互作用蛋白,即激活素受体相互作用蛋白4(ActRIP4)基因。方法:应用ActRIP2基因片段作为探针,从小鼠cDNA文库中钓取ActRIP4 cDNA;采用哺乳细胞双杂交进一步确定ActRIP4与ActRⅡA的相互作用;RT-PCR检测ActRIP4 mRNA在组织中的表达情况。结果:克隆的ActRIP4基因全长691bp,ORF区编码118个氨基酸残基;ActRIP4与ActRⅡA具有特异性结合作用;天然的ActRIP4 mRNA在小鼠多种组织中表达。结论:ActRIP4属于ActRIP家族新成员,可以与ActRⅡA相互作用,可能在多种组织中介导激活素受体信号传导过程。

【Abstract】 Objective To clone the gene of a novel activin receptor-interacting protein 4(ActRIP4) which interacts with type Ⅱ activin receptor(ActRⅡA).Methods The ActRIP4 gene from a mouse cDNA library was cloned by using the ActRIP2 gene fragment as the probe.By mammalian cell two-hybrid,its ability to combine with ActRⅡA was confirmed.The expressions of ActRIP4 mRNA in tissues were analyzed by RT-PCR.Results The cloned full-length cDNA for ActRIP4 was 691bp,which ORF was encoding 118 amino acids.The specific mammalian cell two-hybrid analysis revealed that ActRIP4 could specifically combine with ActRⅡA.The results of RT-PCR showed that ActRIP4 mRNA expressed in many tissues.Conclusion ActRIP4 is a novel member of ActRIP family,and has the ability to specifically combine with ActRⅡA,which may mediate signaling transduction of activin receptor in various tissues.
