【摘要】 目的探讨孕妇外周血中胎儿DNA及瘦素含量与子痫前期发病的关系。方法选择30例子痫前期孕妇为子痫前期组(其中轻度18例,重度12例),B超确定所妊娠的胎儿为男性,于孕20周前、发病时、分娩后6 h抽取外周血;另选30例正常孕妇为对照组,B超确定所妊娠的胎儿为男性,对应子痫前期组时间取血液标本。采用荧光定量PCR及放射免疫方法检测孕妇外周血胎儿DNA浓度及瘦素水平。结果孕20周及发病时胎儿DNA浓度,轻度子痫前期较对照组明显升高,重度子痫前期较轻度者明显升高(F=84.80、96.79,q=11.37～17.30,P<0.05);分娩后6 h胎儿DNA浓度,子痫前期组较对照组明显升高(t′=18.65,P<0.05)。子痫前期发病时瘦素值,轻度子痫前期较对照组明显升高,重度子痫前期较轻度子痫前期明显升高(F=26.46,q=19.23、21.24,P<0.05)。子痫前期病人血液中胎儿DNA与瘦素水平呈正相关关系(r=0.997,P<0.05)。结论孕妇外周血胎儿DNA及瘦素水平与子痫前期发病具有相关性。更多还原

【Abstract】 Objective To study the concentration of fetal DNA and leptin in maternal blood and their relationship with preeclampsia. Methods Thirty gravida with preeclampsia,mild 18 and severe 12,were enrolled in this study.The blood was taken at around 20 weeks of gestation,at the onset,and six hours after parturition.And 30 normal gravida served as controls,their blood was taken accordingly.All of this two groups were confirmed ultrasonographically to have a male fetus.Fetal DNA and leptin in all the pregnant women plasma were measured by fluorescent quantitative polymerase chain reaction(FQ-PCR) and radioimmunoassay(RIA). Results The fetal DNA level of the mild patients at about 20-week gestation and at onset was remarkably higher than that the normal control,and that of the severe ones was higher than that of the mild ones(F=84.80,96.79;q=11.37-17.30;P<0.05).At six hours after dilivery,the fetal DNA level of the patients was significantly higher than that of the control group(t′=18.65,P<0.05).During the attack of preeclampsia,the leptin concentration was significantly higher than that of the normal control,and that of the severe cases was higher than that of the mild ones(F=26.46’q=19.23,21.24;P<0.05).A positive correlativity was observed between the fetal DNA and leptin concentration in the patients(r=0.997,P<0.05). Conclusion The concentration of fetal DNA and leptin in maternal blood closely correlates with preeclampsia.更多还原
