富勒醇/镱(Ⅲ)体系共振光散射法测定核酸(英文)

【摘要】 基于核酸对富勒醇/镱Ⅲ体系共振光散射强度的增强作用,建立了一种新的测定核酸的分析方法.富勒醇的水溶液表现出弱的共振光散射性质,但是当三价镱离子存在时,它的共振光散射强度显著增强,形成了一种稳定的富勒醇/镱Ⅲ共振光散射体系.鱼精DNA的加入使得该体系的共振光散射强度进一步增强.在优化条件下,0 -20.0μg/mL浓度范围内,共振光散射强度的增强值与鱼精DNA的浓度呈线性关系,检出限可达13.3 ng/mL.该法用于合成样品的分析,结果令人满意.

【Abstract】 A sensitive method for the determination of nucleic acids based on the enhancement of resonance light scattering (RLS) of fullerol/ytterbium(Ⅲ) system has been developed. The fullerol showed a weak RLS spectrum in aqueous solution, but its intensity could be enhanced significantly in the presence of ytterbium(Ⅲ) to form a stable RLS system. Moreover, the RLS signal of fullerol/ytterbium(Ⅲ) system could be enhanced further by fish sperm DNA (fs DNA) and the enhanced RLS intensity was proportional to the concentration of fs DNA in the range of 0-20.0 μg/mL with a limit of detection of 13.3 ng/mL under the optimum conditions. The synthetic samples were analyzed with satisfactory results.
