反相高效液相色谱法测定泽兰中乌索酸和齐墩果酸的含量

【摘要】 采用反相高效液相色谱法测定泽兰中乌索酸和齐墩果酸的含量。以NUCLEODUR C18RP柱(250 mm×4.60 mm,5μm)为分析柱,甲醇∶水(88∶12)为流动相,加磷酸调pH值至3.0,流速0.8 ml/min;检测波长为210 nm,柱温25℃。乌索酸进样量在0.368~7.360μg,齐墩果酸进样量在0.200~4.000μg与色谱峰面积呈良好的线性关系。方法准确、快速、灵敏、重现性好,在该色谱条件及样品处理方法下,可以准确测定泽兰中乌索酸和齐墩果酸的含量。

【Abstract】 The determination method of the content of ursolic acid and oleanolic acid in Herba Lycopi with RP-HPLC was established.The HPLC was conducted on a NUCLEODUR C18 RP column(250 mm×4.60 mm,5 μm) with methanol-water solution(88∶12).The solution was adjusted to pH 3.0 with phosphoric acid and used as mobile phase with the flow rate of 0.8 ml/min.The detective wavelength was set to 210 nm and column temperature at 25 ℃.The area under the peak of chromatography was very well correlated with the injected amount of ursolic acid in the range of 0.368~7.360 μg and oleanolic acid in the range of 0.200~4.000 μg.This method was simple and accurate.The experimental results were very reproducible.Under the condition stated in the method section,the determination of ursolic acid and oleanolic acid from Herba Lycopi can be separated very well.The concentration of ursolic acid and oleanolic acid,therefore,can be accurately determined.
