下瘀血汤对进展期大鼠肝纤维化的抑制作用及其方证探讨

【摘要】 目的:探讨下瘀血汤对进展期四氯化碳(CCl4)大鼠肝纤维化的干预作用。方法:采用50%CCl42ml/kg体重皮下注射,每周2次;第9周开始随机分为模型对照组、下瘀血汤组及小柴胡汤组(灌胃给药);用药的同时继续CCl4造模至12周末杀鼠取材。观测肝功能、肝组织病理学、肝组织羟脯氨酸(Hyp)含量变化。结果:8周末模型大鼠肝功能显著异常,肝组织Hyp含量显著增加,肝纤维化明显;12周末模型对照组天冬氨酸氨基转移酶(AST)、碱性磷酸酶(ALP)、γ-L-谷氨酰转肽酶(GGT)、总胆红素(TBil)、Hyp进一步增高,且ALP、GGT、TBil显著高于8周模型对照组(P<0.05)。与12周模型对照组比较,下瘀血汤组丙氨酸氨基转移酶(ALT)、AST、ALP、GGT、TBil、Hyp显著降低(P<0.05或P<0.01)。结论:下瘀血汤能显著抑制造模因素持续刺激条件下进展期肝纤维化的发展,疗效优于小柴胡汤,方证相关有其病理学基础。

【Abstract】 Objective:To probe the interfering action of Xiayuxue Decoction on hepatic fibrosis at progressive stage in rats .Methods:The rats were given subcutaneous injection of 2 ml of 50% carbon tetrachloride(CCl4),twice each week. At the 9th week they were randomly assigned to a model control group,a Xiayuxue Decoction group and a Xiaochaihu Decoction group(administration by intragastrical perfusion).At the same time,CCl4 was continuously given till the end of the 12th week,and the rats were killed for taking samples.Changes of hepatic functions,hepatic histopathology,and hydroxyproline(Hyp) content in the liver were investigated.Results:At the end of 8th week,model rats showed abnormal hepatic function,Hyp content of the hepatic tissue increased significantly with obvious hepatic fibrosis; at the end of 12th week,aspartic aminotransferase(AST),alkaline phosphatase(ALP),r-L-glutamyl transpeptidase(GGT),total bilirubin(TBil),and Hyp increased further in the model control group,ALP,GGT,TBil being significantly higher than those at the 8th week(P<0.05).Compared with the model control group at the 12th week,alanine aminotransferase(ALT),AST,ALP,GGT,TBil,Hyp decreased significantly(P<0.05 or P<0.01).Conclusion:Xiayuxue Decoction can significantly inhibit the development of hepatic fibrosis at the progressive stage under the continuous stimulation of the modeling factors,and its therapeutic effect is superior to that of Xiaochaihu Decoction.The correlativity of prescription with syndrome has pathological basis.
