小柴胡汤治疗子宫内膜异位症的实验研究

【摘要】 目的探讨小柴胡汤对子宫内膜异位症(内异症)大鼠异位内膜的影响及其作用机制。方法将内异症模型大鼠,分为5组,小柴胡汤低剂量组(5g·kg-1·d-1,n=10)、小柴胡汤中剂量组(10g·kg-1·d-1,n=10)、小柴胡汤高剂量组(15g·kg-1·d-1,n=10)、丹那唑组(0.1g·kg-1·d-1,n=9)及模型对照组(20ml·kg-1·d-1蒸馏水,n=10),连续给药4周后,利用透射电镜技术及免疫组化的方法,观察小柴胡汤对内异症大鼠异位内膜的影响;分析异位内膜Fas及Caspase-3蛋白的表达。结果小柴胡汤各组异位内膜生长明显受抑制(P<0.05,P<0.01);异位内膜Fas蛋白、Caspase-3蛋白的表达水平明显高于其在位内膜。结论小柴胡汤能抑制内异症大鼠异位内膜的生长,其作用机制可能是通过上调Fas蛋白的表达,促进异位内膜细胞的凋亡而实现的。

【Abstract】 Objective To explore the effect of Xiaochaihu Decoction(XCHD)on ectopic endometrium in rats with endometriosis and its mechanism.Methods The rat model of endometriosis was established and rats were divided into 5 groups,the low dosage of XCHD group(5g·kg-1·d-1,n=10),the medium dosage of XCHD group(10g·kg-1·d-1,n=10),the high dosage of XCHD group(15g·kg-1·d-1,n=10),the danazol group(0.1g·kg-1·d-1,n=9)and the control group(20 ml·kg-1·d--1 distilled water,n=10).Af- ter treated for four successive weeks,the effect of XCHD on eetopic endometrium was evaluated by observing the changes in transmission electron microscopy.Fas and Caspase-3 protein expression in endometrium and endometri- otic tissue were observed by immunohistoehemistry method.Results Ectopic endometrial growth was markedly in- hibited(P<0.05 or P<0.01)in all the XCHD groups;Fas and Caspase-3 protein expressions of ectopic endom- etrial tissue were higher than those in the endometrium.Conclusion XCHD could inhibit the growth of the eetop- ic endometrium in rats with experimentally induced endometriosis.The therapeutic mechanism may be related to promoting the apoptosis in ectopic endometrial tissue by increasing Fas protein expression.
