骨碎补与雌激素对去卵巢大鼠骨质疏松作用的对照研究

【摘要】 目的研究骨碎补和雌激素对双侧卵巢切除(OVX)大鼠骨质疏松的作用。方法55只雌性大鼠随机分为正常对照组、假手术组、模型组、雌激素组、骨碎补组。用OVX大鼠建立绝经后骨质疏松模型,用骨组织形态计量学方法测量各组大鼠的骨形态学和动力学参数。结果与模型组比较,各组骨小梁体积、骨小梁厚度、骨小梁密度增加,骨小梁间隙及骨小梁表面积和体积之比减少(P<0.05),而骨皮质宽度未见显著性差异;与模型组比较,各组类骨质表面、单标表面、双标表面、组织水平的骨形成速率减少,类骨质成熟时间延长(P<0.05)。雌激素组和骨碎补组各参数间未见显著差异。雌激素组、骨碎补组与正常对照组、假手术组比较,类骨质表面、单标表面、双标表面、组织水平的骨形成速率增加,类骨质成熟时间缩短(P<0.05),其余各项参数未见显著性差异。雌激素组和骨碎补组各参数间未见显著性差异。结论骨碎补与雌激素作用类似,可抑制绝经后高转换型骨质疏松,增加骨小梁骨量和连接性。

【Abstract】 Objective To study the therapeutic effects of Rhizoma Drynariae and estrogen on osteoporosis in ovariectomized(OVX)rats.Methods Fifty-five female rats were randomly divided into 5 groups,the normal con- trol(normal),the sham operated(sham),the model,the estrogen,and the Rhizoma drynariae(RD)groups; ovariectomized rats were used as postmenopausal osteoperosis model.The changes of morphology and dynamic pa- rameters in different groups were determined by bone histomorphometry.Results Compared with the model group, the trabecular volume(TBV/TTV),trabecular thickness(MTPT)and density(MTPD)in the other four groups were significantly increased,while the trabecular template spacing(MTPS)and the ratio of trabecular surface to trabecular volume(TBS/TBV)significantly decreased(P<0.05);and the osteoid surface(TOS),single label surface[Sfract(s)],double label surface[Sfract(d)]and bone formation rate(Svf)also decreased,while oste- oid maturation period(OMP)increased in the latter four groups.No significant difference of cortical width(MCW) was found between these 5 groups.Compared with the normal and sham groups,TOS,Sfract(s),Sfract(d),Svf in the estrogen and RD groups increased significantly,while OMP decreased;no significant difference was found in other parameters.Conclusion Rhizoma Drynariae has the similar effect with estrogen in maintaining normal tra- becular structure and connection by inhibiting the increased bone turnover of pestmenopausal osteoporosis.
