银杏叶提取物预处理对非协调性异种心脏移植的影响

【摘要】 目的观察银杏叶提取物(金钠多)预处理对豚鼠—大鼠非协调性异种心脏移植物的影响并探讨其作用机理。方法采用颈部异位心脏移植模型,分4组:1组(眼镜蛇毒因子预处理,n=10);2组(眼镜蛇毒因子+金钠多预处理,n=5);3组(眼镜蛇毒因子+环孢素A预处理,n=8);4组(眼镜蛇毒因子+环孢素A+金钠多预处理,n=8)。结果各组移植物病理表现均为急性血管性排斥反应(AVR)改变。移植物平均存活时间分别为1组41 h、2组68 h、3组55 h、4组74 h。金钠多预处理的移植物细胞间黏附分子-1 (ICAM-1)表达减少(P<0.05),金钠多联合环孢素A预处理的移植物CD68和CD57表达下调,血红素氧合酶-1 (HO-1)表达上调,凋亡指数降低。结论银杏叶提取物预处理可延长非协调性异种移植物存活时间,与环孢素A伍用可明显减轻异种AVR损害。

【Abstract】 Objective To investigate the effect of ginkgo biloba extract(ginaton)preconditioning on discord- ant cardiac xenografts from guinea pig to rat,and explore its mechanism.Methods Cervical cardiac transplanta- tion model was established in the rats,which were divided into 4 groups:Group 1[cobra venom factor(CVF)pre- treatment,n=10];Group 2(CVF+ginaton,n=5);Group3[cyclosporine(CsA);Group 4(CVF+CsA+gi- naton,n=8].The survival time and histopathology after xenograft were observed and expressions of intercellular ad- hesion molecule-1(ICAM-1),heme oxygenase-1(HO-1),CD68and CD57were detected.Results Pathologic man- ifestion of grafts showed changes of acute vascular rejection(AVR)in all groups.The mean survival time after car- diac xenograft was 41 hrs in Group 1,68 hrs in Group 2,55 hrs in Group 3 and 74 hrs in Group 4.Expression of intercellular adhesion molecule-1(ICAM-1)decreased after ginaton preconditioning(P<0.05).CD68and CD57ex- pressions were down-regulated,HO-1 expression was up-regulated,as well as the apoptotic index(AI)reduced sig- nificantly after ginaton with cyclosporine A preconditioning.Conclusion Ginaton preconditioning can prolong the survival time after discordant xenograft,and significantly alleviate pathological lesion from acute xenograft vascular rejection combined with eyclosporine A.
