双龙胶囊对哮喘大鼠肺组织白三烯类化合物含量的影响
Effect of Shuanglong Capsule on Content of Leukotrienes Compound of Lung Tissues in Asthmatic Rats
【摘要】 目的研究哮喘大鼠肺组织中白三烯B4(LTB4)、白三烯C4(LTC4)及白三烯D4(LTD4)含量的变化及中药复方双龙胶囊对其的影响。方法将SD大鼠随机分成空白组、哮喘模型组、对照组(地塞米松组)、高、中、低3个中药剂量组。用卵蛋白致敏大鼠,以抗原攻击后建立哮喘模型,并用反向高效液相色谱法(RP-HPLC)测定各组大鼠肺组织匀浆中LTB4、LTC4、LTD4含量,并进行比较。结果哮喘大鼠肺组织中LTB4、LTC4、LTD4含量较正常对照组比均有显著升高,地塞米松和双龙胶囊8.27 g/kg,4.13 g/kg均可明显抑制哮喘大鼠肺组织中白三烯的生成(P<0.05)。结论双龙胶囊可明显抑制哮喘大鼠肺组织中炎性介质白三烯的生成,这可能是双龙胶囊用于治疗支气管哮喘的抗炎平喘作用机制之一。
【Abstract】 Objective To observe the changing contents of leukotriene B4 (LTB4),leukotriene C4 (LTC|4), and leukotriene D4 (LTD4) of lung tissue in asthmatic rats,and explore the effect of Shuanglong Capsule (SLC) on it.Methods SD rats were randomly divided into the nomal group,asthmatic model group,Dexamethasone group and the high,middle and low dose SLC groups.All rats except those in the normal group were sensitized by ovalbumin and challenged with the antigen,and the contents of LTB4,LTC4 and LTD4 in lung tissue of all tile groups were measured by reverse phase-high performance liquid chromatography (RP-HPLC) and compared. Results The levels of LTB4,LTC4 and LTD4 of asthmatic rats were significantly higher than those of rats in the normal group.Dexamethasone and SLC at the dose of 8.27 g/kg or 4.13 g/kg could significantly inhibit the pro- duction of leukotrienes of lung tissue in asthmatic rats (P<0.05).Conclusion SLC can significantly inhibit the formation of inflammatory medium LTs of lung tissue in asthmatic rats,it may be one of the key mechanisms of SLC in anti-asthma and anti-inflammatory action.
