脂质的氧化及其对DNA损伤的研究进展

【摘要】 综述了近年来脂质过氧化及其对DNA损伤的研究进展,并且介绍了抗氧化剂对脂质氧化的抑制作用.由自由基引发的脂质过氧化能够产生自由基中间体和亲电的醛类,它们能够诱发疾病,使组织突变及促使机体老化等.过氧化脂质在生物体内积累可以破坏细胞结构和正常的生理功能,对生物体伤害最大的是引发DNA损伤,主要有DNA链断裂和DNA碱基修饰2种形式.抑制脂质过氧化最有效的手段是使用抗氧化剂,它们对预防多种疾病的发生有积极作用.

【Abstract】 Lipid peroxidation is probably the most extensively investigated process induced by free radicals.It can generate free radical intermediates and electrophilic aldehydes as the principal genotoxicants.These toxic substances may lead to diseases,mutagenesis and aging.Peroxide lipid can give rise to functional and morphologic disturbances in cells and develop a number of tissue injuries.Endogenous DNA damage that may contribute significantly to cancer and other genetic diseases linked to lifestyle and dietary factors induced by lipid peroxidation is believed to play a critical role in humans in a variety of pathological conditions.primarily DNA base modification and DNA strand breaks.The incorporation of antioxidants is effectively helpful in retarding the oxidation of lipids.A large body of the literature supports the notion that antioxidants are radioprotectors and play an important role in preventing many human diseases.In this article the current understanding and recent progress in the lipid peroxidation and DNA damage are summarized.A brief overview of the inhibiting effects of antioxidants is introduced.
