枯草杆菌工程菌高密度发酵抗真菌肽的纯化与活性分析
HIGH DENSITY CULTURE OF B.SUBTILIS DB1342(pSC18-66) HARBORING ANTIFUNGAL PEPTIDE CGA18-66 AND PURIFICATION AND BIOACTIVITY ANALYSIS OF PRODUCT
【摘要】 用30 L发酵罐研究了抗真菌肽枯草杆菌工程菌B.subtilisDB1342(pSC18 66)的高密度发酵工艺.通过补料和蔗糖诱导24 h后,工程菌DB1342(pSC18 66)生物量的A600值达到53,目的蛋白分泌量为25 mg/L.发酵液经硫酸铵沉淀浓缩后,过DEAE-Sephadex A 25离子交换柱和Sephadex G 75分子筛层析进行纯化,产物纯度达到90%以上.抑制真菌实验结果显示:纯化产物能抑制白假丝酵母、镰刀菌和链格孢霉菌的生长.
【Abstract】 The high cell density of B.subtilis DB1342(pSC18-66) strain harboring antifungal peptide was cultured in a 30 liter fermentor.The biomass A600 of DB1342(pSC1866) reached 53,the secreted CGA1866 was 25(mg/L) by feed-batch fermentation and sucrose induction for 24 hours.The fermentation supernatant was concentrated with ammonium sulfate,and the expressed product was purified to 90% with DEAE-sephadex A25 anion exchange chromatography and Sephadex G75 gel filtration.The purified product specifically inhibited the proliferation of Candida albican,Alternaria sp.and Fusarium sp.at 4 μmol/L.
