【摘要】 目的：研究genistein对绒毛膜癌细胞形态学、增殖、分化、细胞周期和凋亡的影响。方法：在绒毛膜癌细胞JAR的培养基中加入不同浓度的genistein,每隔24h吸培养上清,测人绒毛膜促性腺激素（HCG）的含量。采用MTT比色法检测不同浓度genistein对JAR细胞增殖的影响。采用HE染色、电镜和流式细胞术检测JAR细胞经genistein处理72h,细胞形态学和超微结构的变化以及genistein对细胞周期和凋亡的影响。结果：Genistein能抑制绒毛膜癌细胞JAR的增殖和诱导凋亡。100nmoI/mL genistein使JAR细胞HCG分泌量增加,并呈时间依赖性；与对照组G₂/M为（17.44±6.1 6）%比较,100 nmoL/m1L genistein使细胞周期阻滞在G₂/M期,G₂/M期比例为（67.14±1.02）%；Annexin-V和PI双染色提示100nmol/L genistein作用JAR细胞72h后,能导致大部分细胞发生晚期凋亡和坏死。结论：Genistein能抑制肿瘤细胞增殖,诱导分化和凋亡,导致细胞周期阻滞。更多还原

【Abstract】 Objective:To study the effects of genistein on morphological changes,proliferation,differentiation,cell cycle and apoptosis of choriocarcinoma JAR cells.Methods:Choriocarcinoma JAR cells were incubated with different concentrations of genistein.Supernatant was removed and tested for HCG every 24 h.The effect of genistein on JAR cell proliferation was meas- ured by MTT assay.The effects of genistein on morphological and ultrastructural changes of JAR cells were observed by HE staining and electron microscope.Cell cycle and apoptosis were analyzed by flow eytometry（FCM）.Results:Genistein inhibited the proliferation and induced the apoptosis of JAR trophoblast tumor cells.Genistein at 100 nmol/mL increased the secretion of human chorionic gonadotropin（HCG）in a time dependent manner.Genistein arrested JAR cells at G₂/M stage compared with control（67.14%±1.02%vs 17.44%±6.16%）.Conclusion:Genistein inhibits proliferation,induces differentiation and apopto- sis,and blocks cell cycle of choriocarcinoma JAR cells.更多还原
