大豆苷元对大鼠心肌缺血再灌注损伤的保护作用
Protective effect of daidzein on myocardial ischemia-reperfusion injury in rats
【摘要】 目的:观察大豆苷元对大鼠心肌缺血再灌注损伤的保护作用。方法:大鼠于结扎冠状动脉左前降支前10m in经舌下静脉注入5.0、2.5 mg/kg大豆苷元溶液1m l/100g,结扎冠状动脉左前降支使心肌缺血40m in后恢复血流并持续120m in复制缺血再灌注损伤模型。测定心肌组织丙二醛和超氧化物歧化酶值,测定血清乳酸脱氢酶、磷酸肌酸激酶值。取心肌作红四氮唑染色测心肌梗死面积。结果:与模型组比较大豆苷元心肌梗死面积明显缩小,血清LDH、CK值降低,血清MDA值减小,SOD值升高,∑-ST值减少,且呈一定剂量依赖性。结论:大豆苷元对大鼠心肌缺血再灌注损伤具有一定的保护作用。
【Abstract】 Objective:To observe the protective effect of(Daidzein,DD) on myocardial ischemia-reperfusion injury in rats.Methods: Rat’s ischemia-reperfusion injury model was established by ligating left anterior descending coronary artery for 40 min followed by reperfusion for 120 min.32 male rats were randomly divided into 4 groups,the sham operation group,the model group and the two DD(5.0,2.5 mg/kg)groups.The two DD groups were respectively given by way of sublingual vein 10 min before ischemia-reperfusion,and to the sham operation group and the model group,saline was given instead.TheⅡcardioelectic lead of ECG was recorded before myocardial ischemia-reperfusion and after lighting 15min,reperfusion 30,60,90,120 min.and analyzed the ∑-ST of Ⅱcardioelectic lead of ECG of each moment to evaluate the protective effect of treatment of DD.The activity of creatine phosphokinase(CK) and lactate dehydrogenase(LDH) in serum,content of malondialdehyde(MDA) and superoxide dismutase(SOD) in myocardial tissue were examined to evaluate the protective effect of DD.To measure the area of myocardial infarction of myocardial ischemia-reperfusion injury,was by The TTC dyeing of cardiac muscle by another 24 rats(the model group,the two DD groups(5.0 mg/kg,2.5 mg/kg),every group had 8 rats.)and by the same methods.Results :The area of acute mycardial infarction was lesser,the value of ∑-ST was reduced,the activity of LDH and CK were lower in the two DD groups than those in the model group.Content of SOD was signifimntly higher and that of MDA was markedly lower than those in the model group.And all the changes mentioned above show some dose-dependent manner.Conclusion:Daidzein have the protective effects on myocardial ichemia-reperfusion injury in rats.
