藏药独一味对肺结核患者Th1/Th2平衡调节作用的研究
The regulation of traditional tibetan herb lamiophlomis rotate on the production of Th1 and Th2 cytokines by peripheral blood mononuclear cells from patients with tuberculosis
【摘要】 目的探讨藏药独一味对肺结核病患者外周血单个核细胞(PBMC)分泌γ-干扰素(IFN-γ)、白细胞介素4(IL-4)的影响。方法葡聚糖-泛影葡胺密度梯度离心法分离20例肺结核病患者和15例健康对照者PBMC。按加入刺激物的不同,将每份标本分为:RPM I1640组、PPD组、PPD+独一味组。加入相应刺激物后培养72h,收集上清液,采用酶联免疫吸附法(ELISA)检测各组培养上清液中IFN-γ、IL-4的水平。结果与PPD组相比,加入独一味能有效促进肺结核病患者及健康对照者PBMC分泌IFN-γ(P<0.05),IL-4水平无差异(P>0.05)。结论独一味通过诱导IFN-γ分泌,调节Th1/Th2平衡,对结核分枝杆菌感染患者发挥保护性的免疫反应。
【Abstract】 Objective To investigate the effect of lamiophlomis rotate(Benth) Kudo on the production of interferon-γ(IFN-γ) and interferon-4(IL-4) by peripheral blood mononuclear cells(PBMC) from patients with tuberculosis.Methods PBMCs were isolated and cultured from 20 patients with pulmonary tuberculosis and 15 healthy persons respectively.According to the different stimulators,every sample was devided into three groups: RPMI1640,PPD,PPD+ Lamiophlomis rotate(Benth) Kudo.After 72 hours,the supernatants of culture PBMC were collected.The levels of IFN-γand IL-4 were measured by ELISA in supernatants.Results Compared with the PPD group,Lamiophlomis rotate(Benth) Kudo increased the production of IFN-γthan those in the healthy controls(P<0.05).The levels of IL-4 were not different between the two groups(P>0.05).Conclusions Lamiophlomis rotate(benth) kudo could regulate the balance between Th1 and Th2 cytokines by inducing IFN-γ production,which suggested lamiophlomis rotate(benth) kudo enhance the patient defense against the infection of tuberculosis.
