脐血瘦素质量浓度与新生儿免疫功能的相关性研究
The study of correlations between the contents of leptin in cord blood and body immune function.
【摘要】 目的探讨脐血瘦素(leptin)质量浓度与新生儿免疫功能的关系及其可能的免疫调控机制。方法2004年4-10月我室对在本校附属医院儿科正常分娩的早产儿34例、足月儿56例,分别采用放射免疫分析(R IA)、免疫透射比浊法及流式细胞术(FCM)检测脐血leptin、粒细胞-巨噬细胞集落刺激因子(GM-CSF)、IgG、IgA、IgM、补体(C3、C4)、白细胞介素-4(IL-4)、r-干扰素(IFN-r)质量浓度及bc l-2蛋白表达量、T淋巴细胞亚群、淋巴细胞凋亡率。结果脐血leptin质量浓度与IgG(r=0.618,P<0.01)、C3(r=0.346,P<0.01)、C4(r=0.272,P<0.05)、CD4+T淋巴细胞百分率(r=0.284,P<0.05)、CD8+T淋巴细胞百分率(r=0.365,P<0.01)及IFN-r质量浓度(r=0.319,P<0.05)、GM-CSF质量浓度(r=0.253,P<0.05)、bc l-2蛋白表达量(r=0.297,P<0.05)均呈正相关,与IL-4质量浓度(r=-0.288,P<0.05)、淋巴细胞凋亡率(r=-0.544,P<0.01)呈负相关,而与IgA、IgM无显著相关(均P>0.05)。结论脐血leptin质量浓度与新生儿免疫功能密切相关,leptin具有增强新生儿细胞免疫功能的作用,而对体液免疫功能影响较小。Leptin可上调bc l-2蛋白表达和GM-CSF质量浓度,抑制淋巴细胞的凋亡,此作用可能是leptin免疫调节作用的重要机制。
【Abstract】 Objective: The main purpose of the study was to research into the correlation of cord blood leptin levels and neonates immunologic function as well as the possible immuno-modulate mechanism.Methods:Thirty-four preterm infants and fifty-six term infants of our hospital were selected from April to October in 2004,respectively.They were checked up the contents of leptin,GMCSF,IgG,IgA,IgM,C3 and C4 in cord blood serum,and the expression of IL-4,IFN-γ,bcl-2 protein,T lymphocyte subpopulation and lymphocyte apoptosis percentage in cord blood by radioimmunoassay(RIA),immuno-transmission turbidity method and flow-cytometry.Results: The umbilical vein blood leptin contents were positively correlated with the IgG(P<0.01),C3(P<0.01),C4(P<0.05) levels,the CD4+ T lymphocyte percentage(P<0.05),CD8+ T lymphocyte percentage(P<0.01),IFN-γ levels(P<0.05),GM-CSF levels and the expression of bcl-2 protein;there had negative correlations between the cord blood leptin levels and IL-4 levels,lymphocyte apoptosis percentage;there had no significant correlation beeween leptin levels and IgA,IgM levels(P>0.05).Conclusion:The cord blood leptin levels are strongly correlated with neonates′ immune functions.Leptin could enhance the cellular immune function.But leptin has minor influence on humoral immunity.Lepin could raise the expression of bcl-2 albumin and the contents of GM-CSF,which is believed to be the possible important mechanisms of the leptin′s immunomodulation function.
