几种表面活性剂的细胞毒性研究
Study on Cytotoxicity of Different Surfactants
【摘要】 目的:研究不同的表面活性剂的细胞毒性,从而筛选出低毒性的表面活性剂,为以后的进一步研究表面活性剂逆转肿瘤细胞的多药耐药提供基础。方法:采用MTT(四甲基偶氮唑盐)检测方法,研究9种不同类型表面活性剂对肿瘤细胞K562/S及其多药耐药株K562/A02的细胞毒性,并比较同一表面活性剂对药物敏感肿瘤细胞和多药耐药肿瘤细胞的细胞毒性之间的差别。结果:非离子表面活性剂对肿瘤细胞K562/S及其多药耐药细胞K562/A02的IC50在10-1mg·ml-1数量级,阳离子及阴离子表面活性剂的IC50为10-3 mg·ml-1数量级。结论:非离子表面活性剂的细胞毒性最小,阴离子表面活性剂次之,阳离子表面活性剂的细胞毒性最大,同一表面活性剂对药物敏感肿瘤细胞及其肿瘤耐药细胞的细胞毒性没有差异,表明表面活性剂对肿瘤细胞多药耐药的逆转不是由其本身的细胞毒性引起的。
【Abstract】 Objective: To investigate the difference of the cytotoxicity between different surfactants. Method; MTT assay was used to study the cytotoxicity of nine surfactants on human leukemic cell line K562/S and its multidrug resistant counterpart K562/A02. Result: The IC50 of the non-ionic surfactants was at the grade of 10-1 rag·ml-1 , The IC50, of the negative ion and the positive ion surfactants was at the grade of 10-3mg·ml-1. Conclusion: The cytotoxicity order of different surfactants is: non-ionic surfactants< negative ion surfactants < positive ion surfactants. The same surfactant has the same cytotoxicity on human leukemic cell line K562/S and its multidrug resistant counterpart K562/A02 were showed.
