镰刀菌毒素丁烯酸内酯对软骨细胞分化的影响及硒的保护效应

【摘要】 目的研究镰刀菌毒素丁烯酸内酯(BUT)对软骨细胞分化的影响及硒(Se)的保护作用。方法体外单层及立体培养软骨细胞,分为对照组(不作任何处理)、加Se0·1μg/ml对照组、BUT0·1μg/ml组、BUT1·0μg/ml组、BUT5·0μg/ml组和BUT1·0μg/ml+Se0·1μg/ml组等6个处理组。采用单克隆、多克隆抗体免疫组织化学法,观察各种处理因素对Ⅱ型胶原(ColⅡ)、X型胶原(ColX)、碱性成纤维细胞生长因子(bFGF)及甲状旁腺激素相关多肽(PTHrP)在培养软骨细胞中表达的影响。结果中、高剂量BUT组Ⅱ型胶原表型表达阳性率明显低于对照组(P<0·05);BUT+Se组Ⅱ型胶原表型表达阳性率明显高于中、高剂量BUT组(P<0·05);低、中、高剂量BUT组培养软骨细胞的bFGF和PTHrP的表达显著增高(P<0·05);各组均未见X型胶原的表型表达。结论BUT可影响软骨细胞Ⅱ型胶原的合成,补硒对BUT损伤软骨细胞有一定的保护作用。

【Abstract】 Objective To study the effect of butenolide(BUT)on cultured chondrocytes differentiation and the possible protective effects of selenium(Se). Methods Ex-vivo cultured chondrocytes were divided into six groups: (1)Control group (without BUT and Se);(2) Se 0.1μg/ml control group;(3) BUT 0.1μg/ml group;(4)BUT 1.0μg/ml group;(5) BUT 5.0μg/ml group; and (6) BUT 1.0μg/ml+Se 0.1μg/ml group. The expression of collagen Ⅱ (ColⅡ),collagen X (ColX),basic fibroblast growth factor (bFGF), and parathyroid hormone-related peptide (PTHrP) in (or around) chondrocytes in all groups were analyzed by immunohistochemistry. Results The expressions of ColⅡ in 1.0μg /ml BUT group and 5.0μg /ml BUT group were significantly lower than those in the control group (P<0.05). The expression of ColⅡ in 1.0μg /ml BUT + Se group were significantly higher than those in the 1.0μg /ml BUT group and 5.0μg /ml BUT group(P<0.05). The expressions of bFGF and PTHrP of BUT groups were significantly higher than those in the Se and control groups(P<0.05). No expression of ColX was observed in all groups. Conclusion BUT can affect the collagen Ⅱ synthesis of the chondrocytes. Selenium supplementation may play a protective role.
