【摘要】 目的观察镇肝息风汤含药血清对血管紧张素Ⅱ(AngⅡ)刺激人脐动脉血管平滑肌细胞(VSMCs)增殖、凋亡的影响。方法制备镇肝息风汤含药血清,采用组织贴块法培养VSMC,并以α-SMactin免疫细胞化学法鉴定。用10-6mol/LAngⅡ刺激VSMC,以MTT法观察含药血清对VSMC增殖的影响;Brdu免疫细胞化学法观察含药血清对VSMCsDNA合成的影响;流式细胞术AnnexinV-FITC法观察含药血清对VSMCs凋亡的影响;并以西药依那普利作为阳性对照药物。结果镇肝息风汤含药血清可抑制AngⅡ诱导的细胞增殖,Brdu标记率增加,促使凋亡率升高。结论镇肝息风汤可通过调节VSMCs增殖-凋亡失衡,从而改善血管重塑。更多还原

【Abstract】 Objective:To investigate the effect of Zhenganxifeng Decoction(ZGXFT,镇肝息风汤 ) on vascular smooth muscle cells proliferation and apoptosis induced by AngⅡ .Methods:Serum containing ZGXFT was produced by the method of serum pharmacology.VSMCs proliferation was observed with the methods of MTT and Brdu immuocytochemistry.VSMCs apoptosis level was examined with the method of flow cytometry.Results:ZGXFT reduced the VSMCS proliferation level which induced by AngⅡ .Compared with the group of VSMCs which was treated with AngⅡ stimulated,VSMCs apoptosis index was increased significantly after ZGXFT treated.Conclusion:ZGXFTcan adjust the apoptosis level of VSMCs induced by AngⅡ .ZGXFT may play a role in cell apoptosis,by which it can inhibit abnormal cell apoptosis which was induce by AngⅡ .更多还原
