豆根管食通口服液对食管癌造模大鼠细胞周期调控因子cyclin D1 CDK4 PCNA的影响

【摘要】 目的:观察豆根管食通口服液对食管癌造模大鼠细胞周期调控因子cyc lin D1、CDK4、PCNA的影响,从细胞分子水平探讨该药的作用机制。方法:选用健康W istar大鼠,除随机分出的空白对照组(正常组)外,其余大鼠均腹部皮下注射甲基戊基亚硝胺(MANA)5mg/kg连续20周,末次注射后将造模大鼠随机分为模型对照组(模型组)、豆根管食通高中低剂量组、西药替加氟组(化疗组),1次/天,4周后停止。应用S-P免疫组化法检测大鼠食管组织cyc lin D1、CDK4、PCNA蛋白的表达。结果:①cyc lin D1、CDK4在食管癌模型组表达率明显提高,与正常组比较有显著性差异(P<0.01)。各药物治疗组cyc lin D1、CDK4表达率均有不同程度的降低,管食通高剂量组和化疗组其阳性率降低更为明显,与模型组比较有统计学意义(P<0.05)。②PCNA阳性指数在食管癌模型组明显升高,与正常组比较有显著性差异(P<0.01)。各药物治疗组PCNA阳性指数均有不同程度的降低,尤以化疗组及管食通高剂量组降低最明显(P<0.01)。结论:豆根管食通口服液通过下调癌基因cyc lin D1、CDK4的表达,阻断细胞由G1期向S期过渡,PCNA阳性指数下降,从而抑制大鼠食管癌细胞生长。

【Abstract】 Objective:To observe the effect of DouGenGuanShiTong(DGGST)oral liquids on the cell cycle regulation factor of modeled esophageal carcinoma rats,explore the function mechanism of the medicine at molecular leve1.Methods:Healthy Wistar rats were treated with MANA 5mg/kg(S.C.once/week)for 20 weeks except control group(the normal).After the last injection,the rats were randomly divided into the following groups:esophageal carcinoma group(the model),DGGST high middle low dose group,the 〖MQ(《MQ1》X14.52,X-W〗Tegafur tablets group(the chemotherapy).Fed them NS or test drug once a day for 4 weeks.Four weeks later,the rats were sacrifi ced to examine the immunohistochemical staining of cyclin D1,CDK4,PCNA were performed using a streptavidin-biotin staining technique.Results:①cyclin D1,CDK4 showed high expression obviously in the model group,compared with the normal group(P<0.01).②PCNA labelling index showed high expression in the model group,compared with the normal group(P<0.01).Conclusion:DGGST has good prevention and treatment effect on esophageal carcinoma by decreasing the expression of the oncogene(cyclin D1,CDK4),and stopping the process of cell cycle from G1 to S phase,PCNA labelling index can be reduced to a lower degree,the growth of esophageal carc inoma cells can be suppressed.
