生骨紫金丹在骨折愈合过程中对TGF-β1和bFGF影响的实验研究

【摘要】 目的:探讨生骨紫金丹的作用机理,为其进一步临床应用提供依据。方法:将W istar大鼠72只造骨折模型后随机分为模型空白组,生骨紫金丹组,阳性对照组。于造模后第2周、3周、4周取局部骨痂制成石蜡切片,以免疫组化技术显示TGF-β1和bFGF,进行图象定量分析。结果:在骨折愈合过程,骨痂中可见TGF-β1和bFGF的阳性表达,灰度测定结果显示在第2周、3周TGF-β1和bFGF的阳性表达强度生骨紫金丹组和阳性对照组与模型空白组有高度显著性差异(P<0.01)。结论:生骨紫金丹能促进TGF-β1和bFGF的表达,促进骨折愈合。

【Abstract】 Objective:To observe the effect and influence of SHENG-GU-ZI-JIN pill(SP)on transforming growth factor beta 1(TGF-β1)and basic fibroblast growth factor(bFGF)in fracture healing process in the rat and to provide the better theoretical basis for more extensive clinical applicationl.Methods:72 Wistar rats were randomly divided into three groups,the control group,〖MQ(《MQ1》X12*2.52(39*2,1)-W〗the Sheng Gu ZiJin pill group(SPG),the Xing Ling Gu Bao group(XG).Fractures were made at middle sments of left femur.At the second,third,fourth weeks after fracture,blood serum and bone specimen were collected from every rat,the SPG and the XG were respectively ingested with SP and XIAN-LING-GU-BAO.Bone specimen were sampled to observe changes of TGF-β1 and bFGF with immunohistochemical studies.Results:Following the formatin of inflammation tissues,in which giant cells,macrophages,mesenchymal cells expressed TGF-β1 and bFGF respectively.Although the expression of TGF-β1 and bFGF was the same among three groups,their content was very different(P<0.05,P<0.01)at second and third week.Conclusion:It is one of the machanism & promoting fracture healing with SP that might partly increase TGF-β1 and bFGF in rats with fracture.
