人工外淋巴液诱导新生豚鼠海马神经干细胞分化的实验研究

【摘要】 目的观察新生豚鼠海马神经干细胞在人工外淋巴液中的分化情况。方法将原代培养的新生豚鼠海马来源的神经干细胞进行体外培养,分别置入含有5%~15%人工外淋巴液培养基使其分化,3周后对分化的细胞进行免疫荧光检测,计数产生表达毛细胞标记蛋白MysionⅦa阳性细胞的比例。结果人工外淋巴液能诱导新生豚鼠海马神经干细胞分化为表达毛细胞特异抗体的细胞。结论神经干细胞在人工外淋巴液中可以存活并分化出表达毛细胞特异抗体的细胞,为神经干细胞的移植内耳治疗提供有力的依据。

【Abstract】 [Objective] To observe the effects of artificial perilymph (APL) on the differentiation of neural stem cells from newborn fetal guine. [Methods] APL were added to DMEM/F12-based medium, the growth and differentiate of NSCs was observed, and the cells were examined by hair cell marker in immunocytofluorescence after 3 weeks. [Results] A total of 3.9%~13.6% of the cells treated with APL culture medium respectively expressed immunoreactivity for Mysion Ⅶa. [Conclusions] Immature neural progenitors possess the potential to differentiate into hair cell phenotypes. Immature neural progenitors may be useful as materials for cell transplantation therapy for replacement of damaged inner ear hair cells.
