【摘要】 目的该研究拟探讨趋化因子IL-8和MCP-1在鼻咽癌病人与正常人血清中表达的情况。方法采用RT-PCR和WestemBlot检测39例鼻咽癌组织、2株鼻咽癌细胞系和17例正常鼻咽黏膜组织中的IL-8和MCP-1表达状况。ELISA检测趋化因子IL-8、MCP-1在鼻咽癌病人与正常人血清中表达的情况。结果在mRNA水平,RT-PCR显示IL-8、MCP-1在鼻咽癌组织表达明显高于正常鼻咽黏膜组织(P<0.05),在蛋白质水平,Westernblot分析发现MCP-1表达明显高于正常鼻咽黏膜组织(P<0.01);Epstein-Barr(EB)病毒阳性的鼻咽癌组织表达IL-8、MCP-1较EB病毒阴性的鼻咽癌组织表达增高,且差异有显著性(P<0.05);ELISA检测鼻咽癌患者血清中IL-8、MCP-1发现,其表达较正常人明显增高(P<0.05),而EB病毒阳性的鼻咽癌患者血清中的IL-8、MCP-1表达水平与EB病毒阴性的鼻咽癌患者血清中的表达水平差异有显著性(P<0.05)。结论趋化因子IL-8、MCP-1在鼻咽癌组织和血清中表达增高,EB病毒感染能促进二者的表达增加。更多还原

【Abstract】 [Objective] Exploited the expressions of chemotacitc factor IL-8 and MCP-1 in the normal people or the patients of nasopharyngeal carcinoma.[Methods] Detected the expressions of chemotacitc factor IL-8 and MCP-1 in 39 case patients of nasopharyngeal carcinoma,two nasopharyngeal carcinoma cell lines and 17 case nasopharyngeal tissues of normal people by RT-PCR and Western Blot.Detected the level of chemotacitc factor IL-8 and MCP-1 in the sera of the naspharyngeal carcinoma patients or normal people by ELISA.[Results] The results show that the expressions of chemotacitc factor IL-8 and MCP-1 in nasopharyngeal carcinoma is higher obviously than in normal nasopharyngeal tissues(P <0.01)in the level of mRNA by RT-PCR.The expressions of chemotacitc factor IL-8 and MCP-1 was significantly different between the patients of nasopharyngeal carcinoma infected by EBVS and the patients of nasopharyngeal carcinoma free EBVS(P <0.05).The detections show that the level of chemotacitc factor IL-8 and MCP-1 in the sera of the naspharyngeal carcinoma patients was significantly higher than in the sera of normal people by ELISA(P <0.05),and the level of chemotacitc factor IL-8 and MCP-1 in the sera was significantly different between the patients of nasopharyngeal carcinoma infected with EBVS and the patients of nasopharyngeal carcinoma free EBVS(P <0.05).[Conclusion] The expression of chemotacitc factor IL-8 and MCP-1 are up-regulated in the tissue or serum of the nasopharyngeal carcinoma patients,which may be involved into the autoimmune protection.The expressions of IL-8 and MCP-1 were relative with EBVS.更多还原
