【摘要】 目的检测c-kitR在急性白血病(acuteleukemia,AL)的表达并进一步分析ANLL中c-kitR表达与染色体异常核型的关系及其意义。方法采用流式细胞术(flowcytometry,FCM)分别检测并比较92例急性淋巴细胞性白血病(ALL)和81例ANLL初诊患者骨髓单个核细胞(BMMNC)跨膜c-kitR表达的阳性率及阳性水平,并设立20例正常人骨髓标本作为阴性对照组;采用R带显带方法对c-kitR阳性表达的ANLL进行染色体核型分析。结果ALL患者的c-kitR表达的阳性率及阳性水平与正常人相比差异无显著性(P>0.05)。ANLL患者的c-kitR表达率及阳性水平则均明显高于正常人和ALL患者(P值分别<0.01)。ANLL中c-kitR阳性表达者异常核型的百分率为74.1%,明显高于c-kitR阴性组者(26.1%)(P<0.05)。异常核型的类型有5q-、-17、+21、del3p、t(8;21)、6p+、t(1;8)、复杂易位、t(15;17)、t(1;12)、-7、四倍体、20q-等十余种类型。结论c-kitR可作为髓系免疫学标记,用以协助ANLL的诊断。c-kitR阳性者伴有细胞遗传学异常,但是,c-kitR阳性与染色体异常核型的类型之间不存在特殊对应关系。c-kit原癌基因不是ANLL发病机制的唯一要素。更多还原

【Abstract】 [Objective] To explore the expression of c-kit receptor(c-kit R,CD117)in acute leukemia(AL).The focus of the study was laid on relationship between expression of c-kit R and abnormal karyotype in ANLL.[Methods] Flow cytometry(FCM)was used to determine the expression of the transmembrane c-kit R on the bone marrow mononuclear cells(BMMNC)in 92 patients of ALL and 81 patients of ANLL.Chromosome R banding technique was used to assay the karyotype in ANLL with positive expression of c-kit R.[Results] There was no significant difference between expression of c-kit in health individuals and patients of ALL(P >0.05).The rate of c-kit R expression in patients of ANLL was significantly higher than that in healthy individuals and that in patients of ALL as well(P < 0.01 respectively).Percentage of abnormal karyotype in the group of positive expression of c-kit R is 74.1%,which was significantly higher than that of 26.1% in the group of negative expression of c-kit R(P <0.05).There were many types of abnormal karyotype in ANLL with positive expression of c-kit R.[Conclusion] c-kit R can be used as the immunology indicator for diagnosis of ANLL.Expression of c-kit R is correlated with abnormal chromosome.But there is no any specific corresponding relationship between the c-kit R expression and the types of abnormal karyotype in ANLL.c-kit oncogene will not be sole premise of pathogenesis of ANLL.更多还原
