大鼠肝缺血再灌注损伤过程中apoM mRNA的表达

【摘要】 目的探讨apoMmRNA在大鼠肝缺血再灌注损伤中的表达。方法建立大鼠肝缺血再灌注损伤模型,健康雄性SD大鼠40只随机分成5组,每组8只:假手术组(对照组);IR1组(肝缺血30min);IR2组(肝缺血60min);IR3组(肝缺血120min);IR4组(肝缺血180min)。IR组的再灌注时间统一为60min。观察每1组的病理变化;检测各组血浆谷丙转氨酶水平(ALT);RealtimeRT-PCR检测apoMmRNA在肝脏中的表达。结果肝缺血再灌注后,光镜下大鼠肝组织有明显的肝血窦和中央静脉瘀血,内皮细胞及肝细胞普遍水肿变性,可见肝细胞坏死,这些变化随着缺血时间的延长而逐渐加重。同样的结果见于血浆中ALT的水平。而apoMmRNA的表达有所不同,与对照组相比,IR1组的表达明显下降(P<0.05),随着缺血时间的延长,apoMmRNA的表达开始上升,至IR4及IR5组,其表达已明显高于IR1组(P<0.05)。结论在肝缺血再灌注损伤过程中,肝内apoMmRNA的表达有迅速、明显的变化,提示apoM可能具有急性时相反应蛋白的特性。

【Abstract】 [Objective] To investigate the expression of apoM mRNA in liver under rat hepatic ischemia reperfusion injury. [Methods] 40 healthy male Spraque Dawley (SD) rats were randomly divided into five groups, i.e., sham-operated group: only hepatoduodenal ligament was separated after entry into the belly, blood flow was not blocked, served as control, and the local ischemia-reperfusion injury (IRI) groups. Local IRI model experienced ischemia for 30~180 minutes followed by reperfusion fro 60 minutes. Serum levels of alanine-aminotransferase (ALT) were determined and small piece of liver tissue from the left lobe was carefully excised for histological analysis. Hepatic apoM mRNA levels were determined by real time RT-PCR method. [Results] Serum ALT levels were significantly elevated in the groups had ischemia reperfusion injury compard to the sham-operated group, i.e., longer ischemia time, higher serum ALT levels. Similar changes were found in the HE staining. ApoM mRNA level was obviously decreased after half hour-ischemia (P <0.05), however, it significantly increased in the groups which had ischemia for 2 or 3 hrs(P <0.05). [Conclusion] ApoM mRNA expression pattern in liver showed a rapid and significant changes during hepatic ischemia-reperfusion injury, which suggests that apoM might have the character of acute phase reactive protein.
