【摘要】 目的:验证无症状乙型肝炎病毒携带者(A SC s)与正常对照组消减文库中的差异基因穿孔素(perforin)在相应人群中的差异表达。方法:采集43例A SC s和41例正常对照者的外周静脉血,分离外周血单个核细胞(PBM C)并提取RNA,做定量RT-PCR(R ea l-tim e RT-PCR)。结果:穿孔素在HBV携带者中表达水平是正常对照的3.58倍。结论:通过定量RT-PCR首次证实了穿孔素基因在A SC s较正常对照表达增高,因穿孔素与免疫抑制有关,该基因在A SC s的高表达对于阐明A SC s的发病机制有重要意义。更多还原

【Abstract】 Objective:To identify the differentially expression of the gene perforin from the subtracted libraries between asymptomatic HBV carriers(ASCs) and the normal control subjects.Methods:2～5 ml venous blood was collected from the subjects of 43 ASCs and 41 the normal control,peripheral blood mononuclear cells(PBMC) were isolated freshly,RNA was extracted from the PBMC for real time RT-PCR.Results:Perforin was confirmed as up expressed by 3.58 fold in ASCs subjects compared to the normal control subjects by real time RT-PCR.Conclusion:Perforin is first confirmed as over-expressed in ASCs compared to the normal control,for perforin is associated with depression of immunity,so the over-expression of perforin in ASCs can play important role in the mechanisms of ASCs更多还原
