当归抗大鼠肺间质纤维化的实验研究

【摘要】 目的探讨当归抗肺间质纤维化的作用机制和应用价值。方法48只大鼠随机分为空白对照组、肺纤维化模型组、氢化可的松治疗组和当归治疗组。气管内注入博莱霉素造成动物模型后,于当天开始每天给药,分别于第7、28d处死,取肺组织进行免疫病理学检查、血浆黏度、全血黏度、血栓素检测。结果转换生长因子β1(TGF-β1)在肺组织中的表达,激素组和当归组均弱于模型组,第7d激素组和当归组无差异,但第28d当归组TGF-β1的表达弱于激素组;模型组全血黏度、血浆黏度、血栓素大多高于空白组;第28d,当归组的全血黏度、血浆黏度低于激素组,而且部分指标低于模型组,激素组则高于模型组;当归组TXB2的含量明显低于激素组和模型组,激素组与模型组无差异。结论当归通过抑制TXB2、血小板活化、TGF-β1,达到抗肺间质纤维化的作用;同时可改善肺间质纤维化形成中的高凝状态而改善肺内微循环,避免激素使用时引起的血液黏度增高,其抗肺间质纤维化作用途径较激素多,某些作用强与激素。

【Abstract】 Objective To observe the inhibitive effect of angelica sinensis on the development of pulmonary fibrosis. Methods 48 Rats were divided into four groups at random: normal control group,untreated animal group,angelica sinensis treated group and hydrocortisone treated group. Fibrosis was induced in rats by intratracheal instillation of bleomycin,then the rats received daily stomach instillation. On days 7 and 28,six rats in each group were killed,lungs and serum were harvested for hisopathological studies,detection for blood viscosity,plasma vascosity and TXB2. Results The expression of TGF-β1 in lung in two treated groups is lower than that of untreated animal group. On day 28 the expression in angelica sinensis treated group is lower than that of hydrocortisone treated group. The values of blood viscosity,plasma vascosity and TXB2 in model group is mostly higher than those in normal control group. On day 28 those values in angelica sinensis treated group is lower than that of hydrocortisone treated group. Conclusion Inhibitive route of angelia sinensis on lung is more than that of hydrocortisone. Some inhibitive effect is stronger than that of hydrocortisone. Angelia sinensis also improves the value of blood viscosity,plasma vascosity,inhibit TXB2 and TGF-β1 expressing in lung,which play an important role in the process of pulmonary fibrosis induced by bleomycin. Angelia sinensis can avoid value of blood viscosity rising which hydrocortisone results in.
