养血清脑颗粒治疗慢性脑缺血衰老大鼠的实验研究

【摘要】 目的观察养血清脑颗粒对衰老大鼠慢性脑缺血的影响,并据此探讨其可能的作用机理。方法将健康雄性Wistar大鼠随机分为空白对照组、模型组、治疗高剂量组、治疗低剂量组和对照组。空白对照组不做任何处理。其余各组大鼠每日用0.5%的D半乳糖(Dgal)生理盐水按50mg/kg颈背部皮下注射,共6周,诱导亚急性衰老大鼠模型;一个月后,永久性结扎大鼠双侧颈总动脉,以复制老年慢性脑缺血模型;治疗高低剂量组和对照组分别给予养血清脑颗粒和尼莫地平片配制的混悬液灌胃;6周后,下腔静脉取血,处死动物,检测血样中的内皮素(ET)、肿瘤坏死因子(TNF)、一氧化氮(NO)、超氧化物歧化酶(SOD)、丙二醛(MDA)。结果治疗高剂量组SOD、NO均升高(P<0.05),ET、TNF、MDA显著降低(P<0.01);治疗低剂量组NO升高(P<0.01),ET、TNF降低(P<0.01,P<0.05);对照组NO升高、TNF降低(P<0.05),其他指标虽有变化,但无统计学意义。结论在Dgal诱导的亚急性衰老大鼠模型的基础上结扎双侧颈总动脉可以制备老年慢性脑缺血模型。养血清脑颗粒可以有效治疗衰老大鼠慢性脑缺血,降低脑缺血后MDA、TNF、ET,升高SOD、NO。

【Abstract】 Objective To observe the effect and investigate the possible mechanism of Yangxueqingnao granule on chronic cerebral circulation insufficiency. Methods Healthy male Wistar rats were randomly divided into 5 groups: blank control group, model group, high-dose therapy group, low-dose therapy group and control group. Induce subacute aging model by D-gal for 6 weeks and ligate the bilateral arteria carotis communis of rats to prepare CCCI model of aging rats. One month later, observe the influence of Yangxueqingnao granule on SOD、MDA、NO、TNF、ET in plasma. Results The plasma SOD, NO were increased(P<0.05)and ET, TNF, MDA were decreased(P<0.01) in high dose therapy group;the plasma NO was increased(P<0.01)and ET, TNF were decreased(P<0.01,P<0.05). NO was increased,TNF was decreased in control group(P<0.05). Conclusion Above method can prepare chronic cerebral circulation insufficiency model of aging rats. Yangxueqingnao granule can treat CCCI effectively.
