益活清胰Ⅰ号对重症急性胰腺炎大鼠胰腺组织缺血的影响

【摘要】 目的观察大鼠重症急性胰腺炎(SAP)时血浆内皮素1(ET1)、血管性假血友病因子(vWF)、6酮前列腺素E1a(6ketoPGE1a)、血栓素B2(TXB2),血小板最大聚集率,胰腺血流量的变化,以及益活清胰Ⅰ号的干预作用。方法81只SD大鼠随机分为假手术组(A组)和SAP模型组(B组),益活清胰Ⅰ号治疗组(C组),每组27只。LSⅢ型组织血流仪检测胰腺的血流量,放免法检测血浆ET1、血清TXB2及6ketoPGE1a含量,酶联免疫吸附法测定血浆vWF含量,全自动血小板聚集仪检测胶原和花生四烯酸诱导的血小板最大聚集率。结果B组大鼠各时点胰组织血流量较A组下降2~5倍(P<0.05),C组各时点胰组织血流量较B组明显改善(P<0.05)。B组大鼠各时点的PAGm和血浆vWF、ET1、TXB2、6ketoPGE1a含量均较A组显著升高(P<0.05),C组大鼠各时点的PAGm和vWF、ET1、TXB2水平均显著低于B组(P<0.05),C组各时点6ketoPGE1a高于B组(P<0.05)。结论益活清胰Ⅰ号具有抑制二十碳烯酸类异常代谢,保护血管内皮细胞,抑制血小板活化和改善微循环的作用。

【Abstract】 Objective To investigate the changes of plasma ET-1 and vWF, 6-keto-PGE_ 1a,TXB_ 2 and pancreatic blood flow in the rats after induction of the severe acute pancreatitis(SAP).The effects of herbs, Yihuoqingyi DecoctionⅠ, on the above index were observed. Methods 81 SD rats were divided randomly into the sham-operated group(group A,n=27), the SAP group (group B,n=27), and the treatment group(group C,n=27). SAP was induced by intraductal administration 5% sodium taurocholate. The abdomen of rats was opened at 6h,12h and 18h after induction of the model for evaluating the pancreatic blood flow by the tissue blood flow instrument. Blood was obtained at 6h,12h and 18h after induction of the model for detecting the concentration of plama ET-1 by ELISA.The concentration of plama vWF and PGE_ 2,TXB_ 2 were detected by radioimmunoassay. The platelet aggregation rate maximum(PAGm) induced by collagen and eicosanoids was detected. Results Pancreatic blood flow of the SAP group was significantly decreased at all time points after induction of the model, compared with those of group A(P<0.05). The contents of plama ET-1, vWF, PGE_ 2 and TXB_ 2 were significantly increased at all time points after induction of the model, compared with those of group A(P<0.05). The PAGm of group B was higher than that of group A(P<0.05). The contents of plama ET-1, vWF, TXB_ 2 and the PAGm were decreased at all time points, compared with those of group B( P<0.05). The PGE_ 2 content of group C was increased, compared with those of group B(P<0.05). Conclusion Yihuoqingyi Decoction Ⅰcan inhibit abnormalities of eicosanoids metabolism and platelet activation, protect endothelial cells, and improve microcirculation.
