酶消化-ICP/MS法测定血中稀土元素的方法研究

【摘要】 目的:建立蛋白酶消化-ICP/MS法同时测定血中13种稀土元素的方法。方法:对蛋白酶K的使用浓度、消化时间、消化温度等条件及ICP/MS仪器测定条件优化,并对血样实测分析。结果:在优化的条件下,蛋白酶K可将血样消化完全,方法操作简单、省时,一次进样可同时测定13种稀土元素,各元素最低检出浓度为0.5~2 ng/L,相对标准偏差均<4.87%,加标回收率96.0%~104%;方法的线性动态范围高达9个数量级,根据血样中各稀土元素含量情况,选用0.005~0.100μg/L为工作曲线范围,相关系数均>0.999。结论:该方法适合于大批量血液样本中13种微量稀土元素的同时检测。

【Abstract】 Objective:To establish a method of enzyme digestion-ICP/MS for the determination of 13 rare earths elements(REE) in blood simultaneously.Methods:The experimental conditions of enzyme digestion such as digestion temperature,digestion time and the analytical conditions of ICP/MS were optimized.Thirteen REE in blood samples were determined by the method of enzyme digestion-ICP/MS.Results:The method enjoyed a wide linear range and good precision,the method detection limit was 0.5~2 ng/L,the average recovery of the method was 96.0%~104%,and the relative standard deviation was lower than 4.87%.Conclusion:The results showed that the method is accurate and highy sensitive and might be good for application and suitable for testing 13 REE in large quantities of blood samples.
