多核白细胞弹性蛋白酶在白细胞介素-1诱导下对角膜基质细胞胶原降解的作用

【摘要】 目的观察多核白细胞弹性蛋白酶及白细胞介素-1α(Interleukin-1α,IL-1α)对人角膜基质细胞胶原降解作用的影响,探讨角膜溃疡的发生机理。方法角膜基质细胞三维胶原凝胶模型表面添加含多核白细胞弹性蛋白酶及IL-1α的MEM液,培养5天,通过测定培养液中的羟脯氨酸(HYP)的量作为胶原降解活性单位。结果多核白细胞弹性蛋白酶直接降解I型胶原,但对角膜基质细胞胶原降解无明显影响,同时添加弹性蛋白酶和IL-1α,明显增加角膜基质细胞胶原降解。结论多核白细胞弹性蛋白酶和IL-1α协同作用促进角膜基质细胞胶原降解,可能在角膜溃疡的发生过程中起着重要的作用。

【Abstract】 Objective To understand the mechanism of corneal ulceration,we investigated the role of neutrophil elastase,interleukin-1 and corneal fibroblasts on collagen degradation.Methods The keratocyte-collagen gels were incubated neutrophil elastase or interleukin-1α at various concentrations were added to the culture medium.Degradation of collagen fibrils during culture was measured by the release of hydroxyproline.Results Neutrophil elastase alone degraded collagens whether absence or presence of corneal fibroblasts and enhanced the collagen degradation by corneal fibroblasts in the presence of interleukin-1.Conclusion Neutrophil elastase not only degraded collagens directly,but also enhanced the keratocytes-mediated collagen degradation stimulated by IL-1α.IL-1αand neuthophil elastase is synergistic effection on contribution to corneal tissue destruction.
