粉红粘帚霉67-1菌株原生质体的形成和再生
Formation and Regeneration of Protoplasts in Gliocladium roseum 67-1
【摘要】 研究了多种细胞壁降解酶的种类和浓度、酶解时间、菌龄,以及渗透压稳定剂等因素对粉红粘帚霉67-1菌株原生质体形成和再生的影响。结果表明,粉红粘帚霉67-1菌株原生质体形成的适宜条件为:使用溶壁酶(3mg/ml)、蜗牛酶(3mg/ml)、纤维素酶(2mg/ml)的组合酶液酶解时间为2h,菌龄为14h,稳渗剂为NaCl(0·5mol/ml)。在上述条件下,原生质体产量达到9·25×106/ml。将原生质体涂布于含KCl(0·5mol/ml)的再生培养基上,再生率最高。显微观察表明,粉红粘帚霉67-1菌株主要通过顶端和侧位释放原生质体。
【Abstract】 The authors studied the effects of lytic enzymes combination, enzymatic time, mycelial age and osmotic stabilizer on the formation and regeneration of Gliocladium roseum 67-1 protoplasts, a highly potential biocontrol agent against plant fungal diseases. Results showed that the optimum conditions for the release of protoplast were enzyme combination of lywallzyme (3 mg/ml), snailase (3 mg/ml) and cellulose (2 mg/ml) with 2 h of enzymatic time, mycelia aged 14 h and NaCl (0.5 mol/ml) as osmotic stabilizer. The optimum osmotic stabilizer for the regeneration of protoplast was KCl (0.5 mol/ml). Microscopical observation indicated that the protoplasts of G. roseum 67-1 were largely released from the apex and the flank of hyphae.
